Tusker FC will kick off their Football Kenya Federation title defense with a tie against AFC Leopards on 25 September 2021.
In a released fixture on Thursday, the season will begin with a Super Cup tie between Tusker FC and Gor Mahia on September 22, 2021, at the Thika Stadium from 3 pm with the match acting as the curtain-raiser for the league.

In week one, Posta Rangers will be up against Kariobangi Sharks with the newly promoted Vihiga Bullets set to host Bidco United at the Bukhungu Stadium. The winner of the FKFPL/NSL PLayoff between Police FC and Vihiga United will travel to Bukhungu on 26th September 2021 to face Kakamega Homeboyz.
